Biological Background: CD38 Function and Localization
- CD38 (ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1) is a multifunctional transmembrane glycoprotein encoded by the Cd38 gene, also known as I-19, ADPRC 1, or Cd38-rs1.
- It functions as both an enzyme and a receptor, catalyzing the synthesis of cyclic ADP-ribose (cADPR) from NAD+, a key second messenger in glucose-induced insulin secretion.
- CD38 also synthesizes nicotinate-adenine dinucleotide phosphate (NAADP+), a potent Ca²⁺ mobilizer, from 2'-phospho-cADPR and nicotinic acid, as well as from NADP+ and nicotinic acid (PubMed:11829748). Related references: PMID:11829748
- The protein possesses cADPR hydrolase activity, thereby regulating intracellular cADPR levels.
- As a receptor, CD38 binds CD31 on endothelial cells, facilitating lymphocyte activation, proliferation, and transendothelial migration.
- In dendritic cells, CD38 signaling is critical for maturation-dependent functions, including CCL21-driven chemotaxis, survival, and Th1-polarizing capacity.
- In lamina propria T lymphocytes, CD38/CD31 interactions trigger LCK and LAT phosphorylation, leading to cytokine release.
- Subcellularly, CD38 is a membrane protein with a single-pass transmembrane domain, and it contains disulfide bonds and glycosylation sites essential for its structural integrity and function.
Experimental Guidance and Technical Tips
- The immunogen covers the extracellular domain (amino acids 45–304), making this antibody suitable for detecting surface CD38 in flow cytometry without permeabilization.
- As a directly conjugated antibody, it is ready for multi-color flow panels; titrate to determine the optimal working concentration for your specific cell type.
- Validate staining in mouse lymphoid tissues or CD38-transfected cell lines; positive controls include activated lymphocytes known to upregulate CD38.
- Cross-reactivity is confirmed for mouse; testing on other species has not been performed and should be validated independently if needed.
