Biological Background: CD366 (TIM-3) Function and Localization

  • CD366 (TIM-3), also known as TIMD-3 or hepatitis A virus cellular receptor 2 homolog, is a type I transmembrane glycoprotein of the immunoglobulin superfamily with a mucin domain, encoded by the Havcr2 gene in mice. It serves as an immune checkpoint receptor with dual inhibitory and stimulatory functions depending on cellular context.
  • TIM-3 negatively regulates Th1-mediated auto- and alloimmune responses, promoting immunological tolerance. Related references: PMID:14556006 PMID:18006747
  • Binding to its ligand galectin-9 (LGALS9) triggers T-cell apoptosis and suppression, mediated by HAVCR2 phosphorylation and BAG6 dissociation, thereby modulating antiviral and antitumor immunity. Related references: PMID:22863785
  • TIM-3 acts as a receptor for phosphatidylserine (PtSer) in a calcium-dependent manner, facilitating phagocytosis of apoptotic cells by dendritic cells and promoting immune tolerance. Related references: PMID:19224762 PMID:20083673
  • In innate immunity, TIM-3 on macrophages regulates activation, and on dendritic cells, it synergizes with Toll-like receptors to enhance TNF secretion. In tumor-infiltrating DCs, it suppresses nucleic acid-mediated innate responses by interfering with HMGB1. Related references: PMID:11823861 PMID:18006747 PMID:22842346
  • TIM-3 is also expressed on natural killer (NK) cells, where it can either co-stimulate IFN-γ production in response to LGALS9 or suppress NK-mediated cytotoxicity, with its negative regulatory role prominent in LPS-induced endotoxic shock. Related references: PMID:25337993
  • Predominantly localized to the cell membrane and cell junctions; a secreted isoform resulting from alternative splicing also exists. The protein undergoes N-linked glycosylation, disulfide bond formation, and phosphorylation during signaling.

Experimental Guidance and Technical Tips

  • As the immunogen covers the extracellular domain (amino acids 20-191), this antibody is expected to detect surface TIM-3 on live, non-permeabilized cells; verify in your sample system.
  • For flow cytometry, optimize the antibody concentration by titration on positive (e.g., activated Th1 cells) and negative (naïve T cells) controls.
  • is a red-excited fluorophore compatible with standard 633–647 nm lasers; ensure proper compensation using single-color controls in multicolor panels.
  • Consider using Fc receptor blocking reagents to reduce nonspecific binding when working with mouse immune cells.
  • TIM-3 expression is dynamic; validate staining under appropriate stimulation conditions (e.g., anti-CD3/CD28 activation) and in your mouse strain of interest.
