Biological Background: CD352/SLAMF6 Function and Localization
- SLAMF6 (signaling lymphocytic activation molecule family member 6), also designated CD352, Ly108, NTB-A, KAL1, and SF2000, is a type I transmembrane receptor belonging to the SLAM family. It contains extracellular immunoglobulin-like domains and cytoplasmic immunoreceptor tyrosine-based switch motifs (ITSMs) that mediate signal transduction. Alternative splicing yields multiple isoforms with distinct functional properties.
- SLAMF6 acts as a self-ligand receptor whose signaling outcomes depend on the presence of small cytoplasmic adapters SH2D1A/SAP and SH2D1B/EAT-2, thereby modulating the activation and differentiation of a wide variety of immune cells in both innate and adaptive immunity. - In natural killer (NK) cells, SLAMF6 triggers cytolytic activity only when high surface densities of natural cytotoxicity receptors are present; this positive signaling involves VAV1 phosphorylation and depends on SH2D1B.
