Biological Background: Hematopoietic Progenitor Cell Antigen CD34 Function and Localization

  • CD34 is a single-pass transmembrane glycoprotein that belongs to the CD34 family; it undergoes alternative splicing to produce multiple isoforms.
  • The protein is thought to mediate stem cell attachment to the bone marrow extracellular matrix or directly to stromal cells, playing a critical role in early hematopoiesis.
  • CD34 presents carbohydrate ligands to selectins, facilitating stem cell binding to lectins expressed by stromal cells or other marrow components (by similarity).
  • It is predominantly localized to the cell membrane.
  • In mouse, CD34 mRNA is highly expressed in hematopoietic progenitor cell lines, brain, and testis, and moderately in thymus, spleen, and bone marrow; it is absent in adult liver (PubMed:1709048). At the protein level, CD34 is detected in the kidney, specifically in the thin limb of Henle’s loop (PubMed:31605441). Related references: PMID:1709048 PMID:31605441
  • Post-translational modifications include glycosylation and phosphorylation, as indicated by the keywords glycoprotein and phosphoprotein.

Experimental Guidance and Technical Tips

  • The immunogen spans the predicted extracellular region (aa 35-287) of mouse CD34, making it suitable for detecting surface-expressed CD34 in flow cytometry without permeabilization.
  • For flow cytometry, titrate the antibody to determine the optimal concentration for your specific cell type and include appropriate isotype controls and viability dyes.
  • Given the expected expression in hematopoietic progenitors, prepare single-cell suspensions from bone marrow or spleen carefully and consider lineage-specific gating strategies.
  • Validate staining in the relevant sample system, as expression levels can vary across tissues and developmental stages.
