Biological Background: Neuropilin-1 Function and Localization

  • Neuropilin-1 (also known as Nrp, NP-1, Npn1, NPN-1, A5 protein) is a single-pass transmembrane receptor encoded by the Nrp1 gene. It has a molecular weight of 103 kDa and belongs to the neuropilin family.
  • It plays a critical role in cardiovascular development and angiogenesis by binding to VEGF165 (isoform of VEGFA) and VEGFB, and enhancing VEGF165 binding to KDR.
  • As a semaphorin receptor, it mediates the chemorepulsant activity of semaphorins, particularly SEMA3A, which is essential for neuronal circuit formation and motor neuron axon guidance during embryonic development. Related references: PMID:26503042
  • It recognizes a C-end rule (CendR) motif R/KXXR/K on ligands, triggering cellular internalization and vascular leakage.
  • In addition to cell membrane localization, Neuropilin-1 is also found in the mitochondrial membrane and cytoplasm, where it regulates mitochondrial iron transport through interaction with ABCB8/MITOSUR.
  • Tissue expression is predominant in the nervous system, consistent with its roles in neurogenesis and neuronal development.
  • Keywords include angiogenesis, calcium, disulfide bond, glycoprotein, heparan sulfate, heparin-binding, and phosphoprotein, reflecting its post-translational modifications and binding properties.

Experimental Guidance and Technical Tips

  • The immunogen covers nearly the full extracellular domain (aa 22-856), suggesting recognition of native extracellular epitopes. Consider validating the antibody for other applications such as immunohistochemistry or immunoprecipitation if proper masking and antigen retrieval are optimized.
  • For flow cytometry, ensure appropriate controls are included to account for potential cell surface shedding or internalization.
  • Since Neuropilin-1 is expressed in the nervous system, consider relevant mouse tissue samples for validation.
