Biological Background: CD24 Function and Localization
- CD24, also known as heat stable antigen (HSA), lymphocyte antigen 52 (Ly-52), and nectadrin, is a small, heavily glycosylated GPI-anchored cell surface protein encoded by the Cd24 gene (Gene ID: 12484).
- It plays a pivotal role in cell differentiation across various cell types, with a specific function in early thymocyte development (by similarity).
- Signaling is triggered by lectin-like ligand binding to CD24 carbohydrates, leading to release of second messengers from its GPI-anchor (by similarity).
- CD24 modulates B-cell activation responses (by similarity).
- In association with SIGLEC10, CD24 is involved in selective suppression of the immune response to danger-associated molecular patterns (DAMPs) such as HMGB1, HSP70, and HSP90 (PubMed:19264983). Related references: PMID:19264983
- CD24 plays a role in the control of autoimmunity (PubMed:20200274). Related references: PMID:20200274
- Subcellular localization: CD24 is anchored to the cell membrane via a GPI-linkage.
- Expressed in lymphoid, myeloid, and erythroid cells.
Experimental Guidance and Technical Tips
- Based on the immunogen (amino acids 27-53), the antibody likely recognizes an epitope in the N-terminal extracellular domain of mouse CD24. Consider this when designing epitope-mapping or competitive binding assays.
- This conjugate is directly fluorescent, simplifying IF/ICC and flow cytometry without secondary reagents. For IF/ICC, optimize fixation and permeabilization to preserve epitope integrity; methanol or paraformaldehyde fixation can be tested.
- For flow cytometry, perform titration to determine the optimal concentration for your cell type; use appropriate isotype controls. Note that CD24 is expressed on lymphoid, myeloid, and erythroid cells, so gating strategies may need to account for broad expression.
- While not listed for Western blotting, the predicted molecular weight is ~8 kDa (core protein), but the mature glycosylated form may appear larger. If WB is attempted, use high-percentage gels and consider glycosidase treatment.
