Biological Background: Signal transducer CD24 Function and Localization
- CD24 is a signal transducer also known as lymphocyte antigen 52 (Ly-52), M1/69-J11D heat stable antigen (HSA), nectadrin, or R13-Ag. It is a small, heavily glycosylated GPI-anchored membrane protein.
- CD24 plays a pivotal role in cell differentiation across various cell types and may have a specific function in early thymocyte development. Signaling is triggered by binding of lectin-like ligands to CD24 carbohydrates and transduced via GPI-anchor-derived second messengers. It modulates B-cell activation responses.
- In association with SIGLEC10, CD24 mediates selective suppression of the immune response to danger-associated molecular patterns (DAMPs) such as HMGB1, HSP70, and HSP90. Related references: PMID:19264983
- CD24 is involved in the control of autoimmunity, as demonstrated in mouse models. Related references: PMID:20200274
- Subcellular localization: CD24 is anchored to the cell membrane via a glycosylphosphatidylinositol (GPI) linkage.
- Tissue specificity: CD24 is expressed in lymphoid, myeloid, and erythroid cells.
- Keywords assigned by UniProt include: Cell membrane, Glycoprotein, GPI-anchor, Immunity, Innate immunity, Lipoprotein, and Signal.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a recombinant fragment spanning amino acids 27-53 of mouse CD24, which covers part of the mature extracellular domain. This region is likely to be well-exposed on the cell surface and suitable for flow cytometry.
- This antibody is recommended for flow cytometry (FC) applications. Titrate the antibody to determine optimal staining conditions for your specific cell types (e.g., mouse lymphoid, myeloid, or erythroid cells).
- Validate the antibody specificity and performance in the relevant sample system, such as mouse splenocytes or thymocytes, using appropriate positive and negative controls.
