Biological Background: Low Affinity Immunoglobulin Epsilon Fc Receptor (CD23) Function and Localization
- CD23, also known as Fc epsilon RII or Fcer2, is a low-affinity receptor for immunoglobulin E (IgE) and the complement receptor CD21.
- Plays critical roles in regulating IgE production and B-cell differentiation; on B cells, it facilitates IgE-dependent antigen uptake and presentation to T cells.
- On macrophages, CD23 engagement triggers intracellular killing of parasites through activation of the L-arginine-nitric oxide pathway.
- The protein exists as membrane-bound and secreted isoforms via alternative splicing.
- Localizes to the cell membrane; a soluble form is also secreted.
- Post-translationally modified with glycosylation, palmitoylation, and proteoglycan additions; contains disulfide bonds and binds calcium.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody spans amino acids 50‐331 of mouse CD23, covering the extracellular lectin domain, which may recognize both membrane-bound and soluble forms.
- For flow cytometry, consider titrating the antibody to determine optimal signal-to-noise ratio for your specific cell type; use appropriate isotype controls.
- Validate reactivity in the relevant mouse sample system to confirm expected staining patterns.
