Biological Background: CD22 Function and Localization
- CD22 is a B-cell receptor and the most highly expressed sialic acid-binding immunoglobulin-like lectin (siglec) on B-cells, playing critical roles in differentiation, antigen presentation, and trafficking to bone marrow (PubMed:7533044, PubMed:9016707). Related references: PMID:7533044, PMID:9016707
- It binds α2,6-linked sialic acid residues on surface molecules such as CD22 itself, CD45, and IgM in a cis configuration, and can also act as an adhesion molecule via trans interactions.
- CD22 functions as an inhibitory coreceptor on B-cells, where it negatively regulates B-cell receptor (BCR) signaling by inhibiting calcium mobilization and cellular activation (PubMed:9016707). Related references: PMID:9016707
- Upon BCR engagement, the immunoreceptor tyrosine-based inhibitory motif (ITIM) of CD22 is phosphorylated by Src kinase LYN, leading to recruitment of protein tyrosine phosphatase PTPN6 (SHP-1) and attenuation of BCR signaling.
- If CD22-mediated negative signaling is sufficiently strong, it can induce apoptosis in B-cells.
- CD22 is localized to the cell membrane and is specifically expressed in B-lymphocytes.
- The protein undergoes alternative splicing, and is classified under keywords including glycoprotein, phosphoprotein, disulfide bond, and immunoglobulin domain-containing protein.
Experimental Guidance and Technical Tips
- This -conjugated antibody is designed for direct detection in immunofluorescence (IF-P) and flow cytometry (FC). For IF-P, consider optimizing fixation and permeabilization steps to preserve membrane localization. A recommended starting dilution for FC is typically between 1:50 and 1:200, but end-users should titrate for their specific experimental system.
- The immunogen corresponds to the extracellular domain (amino acids 22-151), targeting the N-terminal region of mouse CD22. This region includes the V-set Ig-like domain involved in sialic acid binding, suggesting the antibody may recognize the receptor in its native conformation.
- Validate specificity using CD22 knockout or knockdown models, if available, to confirm minimal off-target binding in your assays.
- Since CD22 is a B-cell specific marker, ensure the appropriate cell population is present in your samples. For flow cytometry, include appropriate isotype controls and Fc receptor blocking steps.
