Biological Background: CD19 Function and Localization
- CD19 is a B-lymphocyte surface antigen that functions as a coreceptor for the B-cell antigen receptor complex (BCR), lowering the threshold for activation of downstream signaling pathways and B-cell responses to antigens.
- Upon BCR engagement, CD19 activates phosphatidylinositol 3-kinase and mobilizes intracellular Ca²⁺ stores, amplifying signaling cascades. Related references: PMID:12387743, PMID:20101619, PMID:9382888
- CD19 is not required for early B-cell differentiation in bone marrow but is essential for normal B-1 cell development and for B-cell differentiation and proliferation in response to antigen challenges. Related references: PMID:7542548, PMID:7543183, PMID:9317126
- The protein is required for normal serum immunoglobulin levels and for production of high-affinity antibodies following antigen challenge. Related references: PMID:12387743, PMID:7542548, PMID:7543183
- CD19 is localized to the cell membrane and membrane rafts, consistent with its role in signal transduction.
- Its expression is detected on B cells in spleen, bone marrow, thymus, lymph nodes, and peripheral blood lymphocytes.
- The protein is a glycoprotein with immunoglobulin domains, disulfide bonds, and phosphorylation sites, indicating complex post-translational modification and structural motifs important for its function.
Experimental Guidance and Technical Tips
- is a bright fluorophore; ensure proper compensation and use appropriate controls in multicolor flow cytometry panels.
- Validate the antibody in the relevant mouse B-cell sample system, such as splenocytes, lymph node cells, or peripheral blood lymphocytes.
- The immunogen corresponds to amino acids 19–287, which is the extracellular region of CD19, suggesting the antibody likely recognizes an extracellular epitope suitable for live-cell staining.
