Biological Background: CD150/SLAM Function and Localization

  • CD150/SLAM (SLAMF1) is a self-ligand receptor of the signaling lymphocytic activation molecule family, modulating immune cell activation and differentiation through homo- or heterotypic cell-cell interactions.
  • Two modes of SLAMF1 signaling exist: one depending on SH2D1A/SAP (and perhaps SH2D1B) and another involving PTPN11-dependent signal transduction.
  • In T-cells, SLAMF1 triggers IL-2-independent proliferation and IFN-gamma production, with downstream signaling involving INPP5D, DOK1/2, PRKCQ, BCL10, and NFKB1. Related references: PMID:12351401, PMID:9126961, PMID:11477403, PMID:15539155, PMID:16847311
