Biological Background: CD146 (MUC18) Function and Localization

  • Cell surface glycoprotein MUC18, also known as CD146, melanoma cell adhesion molecule, or gicerin. It belongs to the immunoglobulin superfamily and is a single-pass type I membrane protein.
  • Functions in cell adhesion, contributing to the cohesion of endothelial monolayers at intercellular junctions in vascular tissue. Its expression on melanoma cells may facilitate interactions with vascular elements, enhancing hematogenous tumor spread.
  • During embryonic development, MUC18 may act as an adhesion molecule in neural crest cells.
  • As a surface receptor, it triggers tyrosine phosphorylation of FYN and PTK2/FAK1, and induces a transient increase in intracellular calcium concentration (by similarity).
  • Predominantly localized to the cell membrane; detected in melanoma cell lines.
  • Contains immunoglobulin-like domains, disulfide bonds, and is subject to N-glycosylation and phosphorylation. Alternative splicing generates multiple isoforms.
