Biological Background: CD14 Function and Localization

  • CD14 is a monocyte differentiation antigen and a myeloid-cell-specific leucine-rich glycoprotein.
  • It acts as a coreceptor for bacterial lipopolysaccharide (LPS): in concert with LBP, it binds monomeric LPS and delivers it to the LY96/TLR4 complex, initiating innate immune response. Related references: PMID:16148141
  • LPS-activated CD14 signals through MyD88, TIRAP, and TRAF6, leading to NF-kappa-B activation, cytokine secretion, and inflammatory response. Related references: PMID:15895089, PMID:8612135
  • It also serves as a coreceptor for TLR2:TLR6 and TLR2:TLR1 heterodimers in response to diacylated and triacylated lipopeptides, respectively, triggering signaling from the cell surface and subsequent targeting to the Golgi.
  • CD14 binds electronegative LDL (LDL(-)) and mediates cytokine release induced by LDL(-).
