Biological Background: Interleukin-6 Receptor Subunit Alpha Function and Localization

  • Interleukin-6 receptor subunit alpha (IL-6Rα), also known as IL-6R1 or CD126, is a single-pass type I membrane protein belonging to the type I cytokine receptor family. It contains an immunoglobulin-like domain and requires association with gp130 (IL6ST) for signal transduction.
  • The membrane-bound form of IL-6Rα binds interleukin-6 (IL-6) with low affinity, triggering classic signaling upon gp130 recruitment, which regulates immune responses, acute-phase reactions, and hematopoiesis. This mode is restricted to tissues like the liver and certain immune cells.
  • A soluble form (sIL6R) is generated by shedding or alternative splicing. The IL-6:sIL6R complex (hyper-IL-6) activates gp130 on cells that do not express membrane IL-6R, a process termed trans-signaling that drives pro-inflammatory responses and chronic inflammation (By similarity). It is also required for VEGF production (By similarity) and plays a protective role in liver injury regeneration (PubMed:11113088).
    Related references: PMID:11113088
  • In cluster signaling, membrane-bound IL-6:IL-6R complexes on dendritic cells engage gp130 on neighboring T cells, driving naïve CD4+ T cells toward the Th17 lineage (PubMed:27893700).
    Related references: PMID:27893700
  • IL-6 trans-signaling in the central nervous system regulates energy and glucose homeostasis (PubMed:28402851).
    Related references: PMID:28402851
  • Subcellular localization: IL-6Rα is present on the cell membrane and is also secreted as a soluble receptor. Tissue specificity includes expression by dendritic cells and detection in cerebrospinal fluid.
  • Key post-translational modifications include glycosylation and disulfide bond formation, which are important for its structure and function.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 20–357), making this antibody suitable for detecting both membrane-bound and soluble mouse IL-6Rα in flow cytometry.
  • For flow cytometry, titrate the antibody on relevant cell lines or primary murine cells (e.g., splenocytes, dendritic cells) to determine the optimal staining concentration. Include appropriate positive controls, such as IL-6-stimulated lymphocytes or liver-derived cells.
  • Consider that IL-6R expression is restricted to certain cell types; perform a preliminary expression screen in your sample system to confirm sensitivity.
  • When analyzing soluble receptor dynamics, note that the antibody may also detect IL-6Rα bound to gp130 on adjacent cells in trans-signaling complexes. Validate staining specificity with appropriate blocking controls.
