Biological Background: IL-6 Receptor Subunit Alpha Function and Localization

  • Interleukin-6 receptor subunit alpha (IL-6Rα), also designated CD126, IL-6R, or IL-6RA, is a single-pass type I membrane protein belonging to the immunoglobulin superfamily. It functions as the ligand-binding component of the IL-6 receptor complex, which is essential for IL-6 signal transduction.
  • The membrane-bound IL-6Rα binds IL-6 with low affinity and must associate with the signal-transducing subunit IL6ST/gp130 to initiate intracellular signaling. This classic signaling is restricted to cells expressing IL-6Rα, such as hepatocytes and certain immune cells, and is generally associated with regenerative and anti-inflammatory outcomes. Cluster signaling occurs when membrane-bound IL-6/IL-6Rα complexes on one cell activate IL6ST on adjacent cells.
  • IL-6Rα signaling directs naive CD4+ T cells toward the Th17 lineage through cluster signaling mediated by dendritic cells. Related references: PMID:27893700
  • Soluble IL-6Rα (sIL-6R), generated by proteolytic cleavage or alternative splicing, acts as an agonist of IL-6. The IL-6:sIL-6R complex can stimulate cells that do not express membrane-bound IL-6Rα through trans-signaling, driving pro-inflammatory processes and contributing to chronic inflammatory diseases. Experimental Guidance and Technical Tips

  • The immunogen comprises amino acids 20–357 of the mouse IL-6Rα extracellular domain, which includes the ligand-binding region. This design promotes the generation of antibodies that recognize the native receptor on live cells.
  • For flow cytometry using the conjugate, determine the optimal staining concentration by titration on cells known to express IL-6Rα (e.g., activated T cells, dendritic cells, or hepatocyte lines). Include appropriate isotype and fluorescence-minus-one controls.
  • has excitation/emission peaks similar to Alexa Fluor® 594; avoid spectral overlap with PE, Texas Red, and other red fluorophores. Ensure proper compensation using single-stained controls.
  • Assess cell viability before staining, as dead cells may bind antibodies nonspecifically. Fixation and permeabilization are not recommended for surface antigens unless validated.
  • For blocking, use 5–10% normal serum from the host species of the secondary antibody if indirect staining, but direct conjugate may require minimal blocking. Validate in your system.
