Biological Background: Integrin alpha-M Function and Localization

  • Integrin alpha-M (ITGAM), also known as CD11b, CR3, and MAC-1, is the alpha subunit of the leukocyte integrin heterodimer ITGAM/ITGB2 (CD11b/CD18), a cell surface receptor belonging to the integrin family.
  • The ITGAM/ITGB2 complex functions as a receptor for the iC3b fragment of complement component C3, fibrinogen, and factor X. It mediates adhesive interactions of monocytes, macrophages, and granulocytes, and is critical for the uptake of complement-coated particles and pathogens.
  • This integrin recognizes the R-G-D peptide in C3b and binds to P1 and P2 peptides of the fibrinogen gamma chain. It mediates neutrophil migration and, together with ICAM1, ensures adhesion between stimulated neutrophils and endothelial cells.

  • Subcellularly, integrin alpha-M localizes to the cell membrane and is enriched in membrane rafts, where it facilitates rapid signal transduction.

  • The immunogen comprises amino acids 143–337 of the mouse CD11b extracellular region; this region is likely exposed in the native conformation, making the antibody suitable for detecting cell surface CD11b by flow cytometry.
  • For flow cytometry, titrate the antibody to determine the optimal working concentration and include appropriate isotype controls. Validate staining using mouse monocyte/macrophage cell lines or primary cells such as peritoneal macrophages or bone marrow-derived monocytes.
  • Since CD11b is expressed on granulocytes and microglia, consider using this antibody for immunophenotyping of myeloid cells and for studies involving complement-mediated phagocytosis.
