Biological Background: CD11b (Integrin alpha-M) Function and Localization

  • ITGAM (CD11b) is the integrin alpha-M chain that pairs with integrin beta-2 (ITGB2/CD18) to form the heterodimeric receptor Mac-1 (CR3), a key leukocyte adhesion molecule.
  • It mediates diverse adhesive interactions of monocytes, macrophages, and granulocytes, including binding to complement fragment iC3b (via R-G-D motif), fibrinogen, and factor X.
  • Acts as a receptor for ICAM1, enabling adhesion between stimulated neutrophils and endothelial cells, and regulates neutrophil migration.
  • In microglia, ITGAM cooperates with TYROBP/DAP12 to control superoxide ion production, which promotes neuronal apoptosis during brain development. Related references: PMID:18685038
  • Subcellular localization includes the cell membrane and membrane rafts, consistent with its signaling and adhesion roles.
  • Predominantly expressed in monocytes and granulocytes, with additional expression in a subset of peritoneal mast cells and in microglia at the protein level.
  • Contains disulfide bonds, multiple calcium-binding sites, and a signal peptide; undergoes alternative splicing and is predicted to be glycosylated.

Experimental Guidance and Technical Tips

  • This antibody is directly conjugated to ; users may perform cell surface staining without a secondary antibody. Titrate the antibody to determine the optimal concentration for the specific cell type and application.
  • CD11b is highly expressed on myeloid cells; validate staining in relevant mouse cell lines (e.g., RAW 264.7 macrophages) or primary leukocytes from blood, spleen, or bone marrow.
  • For multicolor flow cytometry panels, include appropriate single-stain controls and compensation controls to correct for spectral overlap, especially with other far-red fluorophores.
  • If intracellular domains are of interest, consider that the immunogen spans amino acids 143–337, an extracellular region; permeabilization protocols should be optimized.
