Biological Background: CD117/c-Kit Function and Localization
- Mast/stem cell growth factor receptor Kit, also known as proto-oncogene c-Kit or tyrosine-protein kinase Kit, is encoded by the Kit (Sl) gene in mouse.
- Acts as a cell-surface receptor for the cytokine KITLG/SCF, playing essential roles in cell survival, proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development and function, and melanogenesis.
- Upon KITLG/SCF binding, activates multiple signaling pathways including PI3K-AKT, RAS-MAPK, and JAK-STAT; phosphorylates PIK3R1, PLCG1, SH2B2, CBL, and recruits GRB2 leading to RAS/RAF/MAPK cascade activation.
- Belongs to the type III receptor tyrosine kinase family, sharing structural features with PDGFR and CSF1R, characterized by immunoglobulin-like domains and a split tyrosine kinase domain.
- Subcellular localization includes the cell membrane and cytoplasm; rapid internalization and degradation upon activation serve to modulate signaling intensity and duration.
- Post-translational modifications include glycosylation, disulfide bond formation, and phosphorylation; alternative splicing generates multiple isoforms with distinct tissue distributions.
- Isoforms 1 and 2 are detected in bone marrow cells, spermatogonia, and spermatocytes; isoform 3 is found in round spermatids, elongating spermatids, spermatozoa, and bone marrow hematopoietic stem cells.
