Biological Background: Megalin/LRP2 Function and Localization

  • Megalin (LRP2) is a type I transmembrane glycoprotein of ~521 kDa, belonging to the low-density lipoprotein receptor family, characterized by multiple ligand-binding repeats, EGF-like domains, and a short cytoplasmic tail.
  • It functions as a multiligand endocytic receptor, mediating the internalization and lysosomal degradation of diverse ligands, including lipoproteins, vitamin-carrier complexes, hormones, enzymes, and drugs, often in partnership with cubilin (CUBN).
  • In the kidney, megalin is critical for tubular reabsorption of filtered proteins; it mediates uptake of metallothionein-bound heavy metals (e.g., cadmium, mercury), the anti-apoptotic protein survivin/BIRC5, vitamin D3/GC-DBP complexes, myoglobin, and matrix metalloproteinases, thereby preserving kidney function.
