Biological Background: Ki67 Function and Localization
- Proliferation marker protein Ki-67 (antigen KI-67, MKI67) is a large nuclear protein (358.7 kDa) that associates with the surface of mitotic chromosomes and plays dual roles as a chromosome repellent during early mitosis and an attractant during late mitosis.
- In early mitosis, Ki-67 forms extended brush structures on chromosomes, preventing their collapse into a single chromatin mass via steric and electrostatic charge barriers, thus enabling independent chromosome motility. Related references: PMID:27362226
- During mitotic anaphase, dephosphorylation of Ki-67 and exposure of a conserved basic patch induce RNA-dependent liquid-liquid phase separation on the chromosome surface, switching it to a chromosome attractant to promote clustering and exclusion of cytoplasmic particles from the future nuclear space.
