Biological Background: IgM Function and Localization

• Isoform 1 of immunoglobulin heavy constant mu (IGHM) constitutes the Fc region of secreted IgM (sIgM). It includes C-terminal segments critical for polymerization and effector interactions.

• Natural sIgM are polyreactive and recognize conserved self- and pathogen-derived structures, while immune sIgM are antigen-specific and induced upon pathogen exposure; both are essential for efficient humoral immunity.

Experimental Guidance and Technical Tips

• The immunogen corresponds to amino acids 218–453 within the constant region of human IgM, suggesting the antibody recognizes a linear epitope conserved in the Fc portion; confirm reactivity on both secreted and membrane–bound IgM in your system.
• For immunofluorescence (IF/ICC and IF-P), permeabilization may not be required to detect surface–expressed IgM, but fixation and permeabilization can be used for intracellular staining if needed.
• In flow cytometry, titrate the antibody to determine the optimal concentration for your cell type, and use appropriate isotype controls and Fc block to minimize nonspecific binding.
• Ensure instrumentation is equipped to detect (excitation/emission maxima ~490/515 nm) and validate the staining pattern with known IgM–positive and –negative cell lines or tissue sections.
