Biological Background: IgD Function and Localization
- Immunoglobulin heavy constant delta (IgD) is the constant region of the delta heavy chain, forming part of immunoglobulins that mediate the effector phase of humoral immunity by eliminating bound antigens.

PMID:22158414 - The variable domains of IgD are assembled through V(D)J recombination and undergo somatic hypermutation to achieve antigen affinity maturation.

PMID:20176268 - Membrane-bound IgD (mIgD) is the major antigen receptor isotype on most peripheral B-cells, coexpressed with IgM, and induces phosphorylation of CD79A and CD79B upon antigen binding.
- Soluble IgD (sIgD) is present in serum at concentrations lower than IgG, IgA, and IgM but higher than IgE; its secretion is shut off following antigen engagement.
- IgD is a potent inducer of cytokines such as TNF, IL1B, IL1RN, IL6, IL10, and LIF from peripheral blood mononuclear cells, with monocytes being the primary producers in vitro.
Related references:
PMID:10702483
PMID:11282392
PMID:8774350 - IgD exists in two forms: membrane-bound on B-cells and secreted into the extracellular space.
- The IgD heavy chain is a glycoprotein with intrachain and interchain disulfide bonds, crucial for its structure and function.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein corresponding to amino acids 99-384 of human IgD, targeting the constant region.
- For flow cytometry, this antibody can be used to detect IgD surface expression on human and cynomolgus B lymphocytes.
- Optimize staining conditions and include appropriate isotype controls.
- Validate the antibody in the specific cell system of interest, as performance may vary with sample preparation and fixation.
- Consider using this antibody in combination with anti-IgM for B-cell subset analysis.
