Biological Background: IgD Function and Localization

  • Immunoglobulin heavy constant delta (IgD) is the constant region of immunoglobulin heavy chains, playing a central role in humoral immunity. It functions both as a membrane-bound B-cell receptor and as a secreted antibody, contributing to antigen recognition and immune response initiation. Related references: PMID:20176268 PMID:22158414
  • The variable region of IgD is formed by V(D)J recombination and can undergo somatic hypermutations, allowing affinity maturation for specific antigens. Related references: PMID:17576170 PMID:20176268
  • IgD is coexpressed with IgM on the surface of most peripheral B cells and serves as the major antigen receptor isotype. Membrane-bound IgD (mIgD) induces signaling via CD79A and CD79B phosphorylation by Src family kinases.
  • Soluble IgD (sIgD) is present in lower serum concentrations than IgG, IgA, and IgM and is shut off after antigen binding.
  • IgD exhibits immunomodulatory properties by inducing the release of cytokines such as TNF, IL1B, IL1RN, IL6, IL10, and LIF from peripheral blood mononuclear cells, with monocytes as the primary producers. Related references: PMID:10702483 PMID:11282392 PMID:8774350
  • IgD exists in membrane-bound and secreted isoforms resulting from alternative splicing and is localized both at the cell membrane and secreted.
  • Post-translational modifications include glycosylation and disulfide bond formation, which stabilize its immunoglobulin domain structure.

Experimental Guidance and Technical Tips

  • The immunogen, covering amino acids 99–384 of human IgD, corresponds to the constant region, ensuring specific recognition of the IgD heavy chain regardless of variable region diversity.
  • The conjugate is optimized for flow cytometry (FC); titrate the antibody to determine the optimal working concentration for your specific cell system.
  • Consider using this antibody to analyze IgD expression on B cells from human and rhesus peripheral blood mononuclear cells (PBMCs) or lymphoid tissues.
  • Co-staining with an anti-IgM antibody can help delineate mature, naive B cells that co-express IgD and IgM.
  • Protect the antibody from light during storage and handling to preserve the fluorophore's stability.
