- CD8A encodes the alpha chain of the T-cell surface glycoprotein CD8, also known as T8/Leu-2, a coreceptor critical for adaptive immunity.
- Functions as a coreceptor for MHC class I molecules, interacting simultaneously with the T-cell receptor (TCR) and MHC class I proteins on antigen-presenting cells to facilitate recognition of cytosolic antigens.
- Recruits Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling cascades that lead to T-cell activation, lymphokine production, motility, adhesion, and cytotoxic T-lymphocyte (CTL)-mediated killing of infected and tumor cells.
- In NK cells, CD8A homodimers provide a survival mechanism enabling conjugation and lysis of multiple target cells, and also promote differentiation of activated lymphocytes into memory CD8 T-cells.
