Biological Background: CD8a Function and Localization

  • CD8a is a T-cell surface glycoprotein, also known as T-lymphocyte differentiation antigen T8/Leu-2, encoded by the CD8A gene.
  • It plays a critical role in adaptive immunity as a coreceptor for MHC class I molecules, interacting with the T-cell receptor (TCR) and MHC I-peptide complexes on antigen-presenting cells.
  • The interaction recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling that leads to cytotoxic T-lymphocyte activation, proliferation, and target cell killing.
  • In natural killer (NK) cells, CD8A homodimers promote survival and enhance conjugation and lysis of target cells; homodimers also support differentiation of activated lymphocytes into memory CD8 T-cells.
  • The protein contains an immunoglobulin domain and undergoes post-translational modifications including disulfide bond formation, glycosylation, and palmitoylation.
  • CD8A is predominantly expressed at the cell membrane of thymus-derived T-cells, usually as a heterodimer with CD8B, but homodimers are found on NK cells, memory T-cells, intraepithelial lymphocytes, monocytes, and dendritic cells.
  • Expressed on plasmacytoid dendritic cells upon herpes simplex virus-1 stimulation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (aa 22-182), targeting an exposed epitope suitable for live-cell flow cytometry.
  • Titrate the antibody to determine the optimal concentration for your specific cell population and staining conditions.
  • As a direct conjugate with , it is compatible with multicolor flow cytometry panels; consider compensation controls and spectral overlap.
  • Validate performance in your experimental system using appropriate positive (e.g., human PBMCs) and negative controls.
