- CD8a is a T-cell surface glycoprotein, also known as T-lymphocyte differentiation antigen T8/Leu-2, encoded by the CD8A gene.
- It plays a critical role in adaptive immunity as a coreceptor for MHC class I molecules, interacting with the T-cell receptor (TCR) and MHC I-peptide complexes on antigen-presenting cells.
- The interaction recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling that leads to cytotoxic T-lymphocyte activation, proliferation, and target cell killing.
- In natural killer (NK) cells, CD8A homodimers promote survival and enhance conjugation and lysis of target cells; homodimers also support differentiation of activated lymphocytes into memory CD8 T-cells.
- The protein contains an immunoglobulin domain and undergoes post-translational modifications including disulfide bond formation, glycosylation, and palmitoylation.
