Biological Background: CD69 Function and Localization

  • CD69 (Early activation antigen CD69; also known as AIM, EA1, CLEC2C, GP32/28, Leu-23) is a C-type lectin transmembrane protein encoded by the CD69 gene.
  • It plays a critical role in immune cell homeostasis, as it is rapidly expressed on the surface of platelets, T-lymphocytes, and NK cells upon activation by antigens or cytokines, stimulating different signaling pathways in various cell types (PubMed:24752896, PubMed:26296369, PubMed:35930205). Related references: PMID:24752896 PMID:26296369 PMID:35930205
  • CD69 negatively regulates Th17 cell differentiation via carbohydrate-dependent interaction with galectin-1/LGALS1 on immature dendritic cells (PubMed:24752896). Related references: PMID:24752896
  • CD69 promotes the conversion of naive CD4 T-cells into regulatory T-cells via the S100A8/S100A9 complex present on PBMCs (PubMed:26296369). Related references: PMID:26296369
  • Acting as an oxidized low-density lipoprotein (oxLDL) receptor on CD4 T-lymphocytes, CD69 negatively regulates inflammation by inducing PDCD1 expression through NFAT activation (PubMed:35930205). Related references: PMID:35930205
  • CD69 participates in protection against Pseudomonas aeruginosa infection by recognizing the pathogen and promoting ERK1 activation, leading to GM-CSF and inflammatory cytokine secretion (PubMed:34841721). Related references: PMID:34841721
  • CD69 negatively regulates chemotactic responses of effector lymphocytes and dendritic cells to sphingosine 1-phosphate (S1P) by acting as an S1PR1 agonist and facilitating receptor internalization and degradation (PubMed:37039481). Related references: PMID:37039481
  • Subcellularly, CD69 is a cell membrane protein; it is expressed on activated T-cells, B-cells, NK cells, neutrophils, eosinophils, epidermal Langerhans cells, and platelets. The protein is a glycoprotein, phosphoprotein, and contains a disulfide bond, with a molecular weight of approximately 22.6 kDa (UniProt: Q07108).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular region (aa 78–199) of human CD69, targeting the surface-exposed domain for flow cytometry detection.
  • As a directly conjugated antibody, it enables one-step staining, reducing background and simplifying multicolor panel design.
  • When implementing flow cytometry assays, consider titrating the antibody to determine the optimal concentration for your specific cell type and activation conditions.
  • Include appropriate isotype controls and compensation controls to ensure specificity and accurate gating.
