Biological Background: CD64 (FcγRI) Function and Localization
- CD64 (FcγRI) is a high-affinity IgG Fc receptor encoded by the FCGR1A gene (also known as FCG1, FCGR1, IGFR1). It belongs to the immunoglobulin superfamily and contains three Ig-like domains.
- Functions as a key mediator of both innate and adaptive immune responses, primarily by binding the Fc region of IgG and triggering antibody-dependent cellular cytotoxicity (ADCC) of virus-infected cells on monocytes/macrophages.
- Specifically expressed on monocytes and macrophages (monocyte/macrophage specific).
- Localized to the cell membrane as a single-pass type I membrane protein.
- Post-translationally modified by glycosylation and phosphorylation; annotated as a glycoprotein and phosphoprotein in UniProt.
- Subject to alternative splicing, generating multiple transcript variants.
- The unmodified protein has a calculated molecular weight of ~42.6 kDa; the product's observed band at 43 kDa likely reflects glycosylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 16-288), making this antibody suitable for detecting surface-expressed CD64 by flow cytometry.
- Ensure proper titration for flow cytometry assays. Since CD64 is monocyte/macrophage specific, include appropriate cell lineage markers to gate on these populations.
- The conjugate provides a bright and photostable signal, suitable for multiplex panels. Validate in the relevant sample system, such as human PBMCs or cynomolgus samples.
