Biological Background: CD45 Function and Localization
- CD45, also known as leukocyte common antigen (L-CA), is a receptor-type tyrosine-protein phosphatase encoded by the PTPRC gene in humans.
- It is a transmembrane glycoprotein expressed on all nucleated hematopoietic cells and exists in several isoforms generated by alternative splicing, resulting in molecular weights ranging from 131 kDa to 147 kDa.
- CD45 plays a critical role in T-cell activation by dephosphorylating key signaling molecules such as LYN, FYN, and SKAP1, thereby modulating T-cell receptor signaling thresholds. Related references: PMID:35767951
- The protein contains two intracellular phosphatase domains; the first (D1) is enzymatically active, while the second (D2) regulates substrate specificity.
- CD45 localizes to the cell membrane and is enriched in membrane rafts at the immunological synapse during antigen-presenting cell–T cell interactions.
- It acts as a receptor for human cytomegalovirus protein UL11, which impairs T-cell proliferation upon binding.
