Biological Background: CD45 Function and Localization

  • CD45, encoded by the PTPRC gene, is a receptor-type tyrosine-protein phosphatase (PTP) also known as leukocyte common antigen (L-CA) or T200. Multiple protein isoforms are generated through alternative splicing.
  • CD45 is required for T-cell activation through the antigen receptor and acts as a positive regulator of T-cell coactivation upon binding to DPP4. Related references: PMID:35767951
  • The PTP possesses two tandem phosphatase domains: the first has catalytic activity, while the second modulates substrate specificity. It dephosphorylates key signaling proteins including SKAP1, FYN, and LYN.
  • At the immunological synapse, CD45 interacts with CLEC10A on dendritic cells, recognizing Tn antigen-bearing CD45 on effector T cells to tune T-cell activation thresholds and limit autoreactivity.
  • CD45 localizes to the cell membrane, membrane rafts, and synapse, consistent with its role in immune signaling.
  • During cytomegalovirus infection, CD45 acts as a receptor for viral protein UL11, leading to impaired T-cell proliferation upon receptor stimulation.
  • In thymocytes, distinct CD45 isoforms exhibit differential expression: isoforms 1, 2, 3, 5, and 7 are detected, while isoforms 4, 6, and 8 are absent.

  • The immunogen comprises the extracellular region (aa 26–577) of human CD45, covering the majority of the ectodomain and making this antibody suitable for detecting all CD45 isoforms in flow cytometry.
  • Given the high and ubiquitous expression of CD45 on leukocytes, proper gating strategies should be employed to distinguish lymphocyte populations. Isotype and fluorescence-minus-one controls are recommended to set appropriate gates.
  • Titrate the antibody to determine the optimal staining concentration for your specific sample type and experimental conditions, ensuring adequate signal-to-noise ratio.
