Biological Background: CD4 Function and Localization
- CD4 is a 51 kDa type I transmembrane glycoprotein belonging to the immunoglobulin superfamily, expressed on the surface of T-helper cells, macrophages, dendritic cells, and NK cells as a hallmark of helper T lymphocytes.
- As a coreceptor, CD4 binds to MHC class II molecules on antigen-presenting cells, simultaneously interacting with the T-cell receptor (TCR) to recruit the Src kinase LCK, which initiates signaling cascades leading to T-cell activation, cytokine production, and differentiation.
- In the thymus, CD4 participates in the positive selection and lineage commitment of CD4+ T-helper cells, essential for adaptive immunity.

- Whole blood staining or staining of lymphoid tissue single-cell suspensions may be considered, following standard protocols with appropriate anticoagulants and viability dyes.
