  • CD4, also known as T-cell surface antigen T4/Leu-3, is a T-cell surface glycoprotein encoded by the CD4 gene, belonging to the immunoglobulin superfamily.
  • Functions as a coreceptor for MHC class II molecules in T-cells, interacting with the T-cell receptor (TCR) and MHC class II on antigen-presenting cells, and recruiting the Src kinase LCK to trigger intracellular signaling pathways leading to T-helper cell activation, lymphokine production, motility, and adhesion.
  • Participates in T-helper cell development in the thymus and triggers the differentiation of monocytes into functional mature macrophages.
  • In macrophages and NK cells, CD4 mediates differentiation, activation, cytokine expression, and cell migration through a TCR/LCK-independent pathway.
  • Localizes to the cell membrane as a single-pass type I membrane protein.
  • Highly expressed in T-helper cells, and also present in other immune cells including macrophages, dendritic cells, and NK cells.
  • Post-translational modifications include disulfide bonds, glycosylation, palmitoylation, and phosphorylation; structural features include immunoglobulin domains, a signal peptide, and a transmembrane helix.
