- CD4 is a 51 kDa type I transmembrane glycoprotein belonging to the immunoglobulin superfamily, also known as T4 or Leu-3.
- It functions as a coreceptor for MHC class II molecules, binding simultaneously with the T-cell receptor (TCR) and MHC class II on antigen-presenting cells, leading to LCK recruitment and downstream signaling.
- CD4 is essential for T-helper cell development in the thymus and drives monocyte-to-macrophage differentiation in a TCR/LCK-independent manner.
- Highly expressed on T-helper cells, CD4 is also present on macrophages, dendritic cells, and NK cells, mediating adhesion and migration.
