Biological Background: CD4 Function and Localization
- CD4 is a T-cell surface glycoprotein, also known as T4/Leu-3, belonging to the immunoglobulin superfamily. It functions primarily as a coreceptor for MHC class II molecules on antigen-presenting cells.
- In T-helper cells, CD4 interacts simultaneously with the T-cell receptor (TCR) and MHC class II:peptide complex, recruiting the Src kinase LCK to initiate signaling cascades that lead to T-cell activation, lymphokine production, and immune response.
- CD4 is highly expressed on the surface of T-helper cells, but is also present on other immune cells such as macrophages, dendritic cells, and NK cells, where it may modulate differentiation and cytokine expression.
- Subcellularly, CD4 is an integral membrane protein localized to the cell membrane, with a single transmembrane helix.
- CD4 serves as the primary receptor for human immunodeficiency virus-1 (HIV-1), facilitating viral entry into host cells.
