Biological Background: CD38 Function and Localization

  • CD38 is a multifunctional transmembrane glycoprotein that functions as both an enzyme and a receptor. It synthesizes cyclic ADP-ribose (cADPR) from NAD+, a key second messenger for glucose-induced insulin secretion, and also hydrolyzes cADPR to ADP-ribose. Related references: PMID:7961800 PMID:8253715

  • It catalyzes the formation of nicotinate-adenine dinucleotide phosphate (NAADP+), a Ca2+ mobilizer, preferentially from 2'-phospho-cADPR and nicotinic acid. Related references: PMID:16690024

  • As a receptor, CD38 binds to CD31 (PECAM-1) on endothelial cells, facilitating lymphocyte activation, proliferation, and transendothelial migration. Related references: PMID:9551996

  • In mature dendritic cells, CD38/CD31 interactions regulate CCL21-driven migration, survival, and Th1-polarizing capacity. Related references: PMID:16293598

  • CD38 engagement in lamina propria T lymphocytes triggers LCK and LAT activation and subsequent cytokine release. Related references: PMID:11259373

  • CD38 is expressed on the cell surface and membrane.

  • It shows high expression in pancreas, liver, kidney, brain, testis, ovary, placenta, malignant lymphoma, and neuroblastoma.

  • The protein undergoes alternative splicing and contains disulfide bonds, glycosylation sites, and a signal-anchor transmembrane domain.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 43–300 of human CD38, covering the extracellular domain, so this antibody is suitable for detecting surface CD38 by flow cytometry.
  • Since the antibody is directly conjugated to , no secondary antibody is needed; consider titrating to determine the optimal staining concentration for your cell type.
  • Cross-reactivity with cynomolgus CD38 has been demonstrated; therefore, the antibody may be used in non-human primate studies, but validation in your specific assay system is recommended.
  • CD38 is expressed on various immune cells (e.g., lymphocytes, dendritic cells) and some tissues; include appropriate isotype controls and cell lineage markers to confirm specificity.
  • The protein is reported to have multiple molecular weight forms (14 kDa and 34 kDa), likely due to glycosylation or isoforms; the observed band pattern may vary depending on the biological context.
