Biological Background: CD38 Function and Localization
- CD38 is a multifunctional transmembrane glycoprotein belonging to the ADP-ribosyl cyclase family, encoded by the CD38 gene (GeneID: 952). It is also known as ADPRC1, cADPR1, and T10.
- It catalyzes the synthesis of cyclic ADP-ribose (cADPR) from NAD+, a second messenger that mobilizes calcium and regulates glucose-induced insulin secretion. Related references: PMID:7961800, PMID:8253715
- CD38 also synthesizes nicotinate-adenine dinucleotide phosphate (NAADP+), another potent Ca2+ mobilizer, and exhibits cADPR hydrolase activity. Related references: PMID:16690024, PMID:7961800, PMID:8253715
- As a receptor, CD38 binds platelet endothelial cell adhesion molecule (CD31) on endothelial cells, promoting lymphocyte activation, proliferation, and transendothelial migration. Related references: PMID:9551996
- In dendritic cells, CD38 regulates maturation-associated functions such as CCL21-driven migration, survival, and Th1-polarizing activity. Related references: PMID:16293598
- CD38/CD31 interactions in lamina propria T lymphocytes trigger LCK and LAT signaling leading to cytokine release. Related references: PMID:11259373
- CD38 is predominantly localized to the cell surface and cell membrane.
- High expression is observed in pancreas, liver, kidney, brain, testis, ovary, placenta, malignant lymphoma, and neuroblastoma.
- The protein contains a signal-anchor, several disulfide bonds, and glycosylation sites, and alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (amino acids 43–300), ensuring reactivity with native CD38 on live cells.
- Direct conjugation to makes this antibody ideal for multi-color flow cytometry; include appropriate compensation controls.
- Validate the optimal antibody dilution and staining protocol in your experimental system, as conditions may vary.
- With confirmed cross-reactivity to cynomolgus monkey, it may be suitable for non-human primate studies.
- Use isotype controls and Fc receptor blocking to minimize non-specific binding.
