Biological Background: CD38 Function and Localization
- CD38, encoded by the CD38 gene, is a multifunctional transmembrane glycoprotein that acts as both an ADP-ribosyl cyclase and a cyclic ADP-ribose hydrolase. Alternative splicing generates isoforms, including a ~34 kDa full-length and a ~14 kDa soluble form, consistent with the observed protein weight range.
- It synthesizes cyclic ADP-ribose (cADPR), a second messenger involved in glucose-induced insulin secretion, from NAD+ (PubMed:7961800; PubMed:8253715).
Related references: PMID:7961800 PMID:8253715 - CD38 also catalyzes the production of nicotinate-adenine dinucleotide phosphate (NAADP+), a potent intracellular Ca2+ mobilizer, from 2'-phospho-cADPR and nicotinic acid, or from NADP+ and nicotinic acid (PubMed:16690024).
Related references: PMID:16690024 - As a hydrolase, CD38 degrades cADPR to ADP-ribose, providing a mechanism to regulate second messenger levels (PubMed:7961800; PubMed:8253715).
Related references: PMID:7961800 PMID:8253715 - Beyond its enzymatic roles, CD38 serves as a receptor for CD31 on endothelial cells, facilitating lymphocyte adhesion, activation, and transendothelial migration (PubMed:9551996).
Related references: PMID:9551996 - In dendritic cells, CD38 regulates maturation-associated processes including CCL21-driven migration, survival, and Th1-polarizing activity (PubMed:16293598).
Related references: PMID:16293598 - CD38/CD31 cognate interactions in lamina propria T lymphocytes activate LCK and LAT, leading to cytokine secretion (PubMed:11259373).
Related references: PMID:11259373 - Localized at the cell surface and membrane, CD38 features a signal-anchor and a single transmembrane helix, consistent with its transmembrane topology. Post-translational modifications include glycosylation and disulfide bond formation.
- Highly expressed in tissues such as pancreas, liver, kidney, brain, testis, ovary, placenta, and in malignant lymphoma and neuroblastoma, CD38 is a target in diabetes mellitus and cancer research.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 43–300), making this antibody well-suited for detecting surface CD38 by flow cytometry.
- is a bright, photostable fluorophore equivalent to Alexa Fluor® 488, enabling direct detection without the need for a secondary antibody.
- Perform antibody titration to determine the optimal concentration for your specific cell type and experimental setup; a typical starting range is 0.5–5 μg per million cells.
- Include appropriate isotype control and viability staining to set gates and exclude non-specific binding.
- Because the antibody cross-reacts with cynomolgus monkey, it can be used in non-human primate studies; however, validation in the relevant sample matrix is recommended.
- For optimal performance, protect the antibody from light and store according to the manufacturer's instructions.
