Biological Background: CD18 Function and Localization
- CD18 encodes integrin beta-2 (ITGB2), a cell surface glycoprotein that pairs with multiple alpha subunits (CD11a-d) to form leukocyte-specific integrins: LFA-1 (CD11a/CD18), Mac-1 (CD11b/CD18), p150,95 (CD11c/CD18), and alphaD/beta2 (CD11d/CD18). These heterodimers mediate diverse cell–cell and cell–matrix interactions.
- As part of LFA-1 (ITGAL:ITGB2), it serves as a receptor for ICAM1, ICAM2, and ICAM3, mediating T-cell adhesion, immunological synapse formation, and leukocyte transmigration across endothelia (PubMed:1676048, PubMed:23775590, PubMed:38195629; PubMed:11812992, PubMed:28807980). LFA-1 also binds the ubiquitin-like protein ISG15 to modulate immune responses (PubMed:29100055).
