Biological Background: CD14 Function and Localization

  • CD14, also designated My23 antigen, is a glycosylphosphatidylinositol (GPI)-anchored glycoprotein and a member of the leucine-rich repeat (LRR) family.
  • Functions as a coreceptor for bacterial lipopolysaccharide (LPS). In concert with LBP, CD14 captures monomeric LPS and transfers it to the LY96/TLR4 complex, triggering innate immune signaling. Related references: PMID:1698311 PMID:23264655
  • Downstream signaling requires the adaptors MyD88, TIRAP, and TRAF6, ultimately leading to NF-kappa-B activation and the secretion of proinflammatory cytokines. Related references: PMID:8612135
  • CD14 also functions as a coreceptor for TLR2:TLR6 and TLR2:TLR1 heterodimers, recognizing diacylated and triacylated lipopeptides respectively.
