Biological Background: CD14 Function and Localization

  • CD14 is a myeloid cell-specific leucine-rich glycoprotein, also known as monocyte differentiation antigen CD14 or My23 antigen.
  • CD14 functions as a key coreceptor for bacterial lipopolysaccharide (LPS), mediating innate immune recognition and response. Related references: PMID:1698311, PMID:23264655
  • In concert with LBP, CD14 binds monomeric LPS and delivers it to the LY96/TLR4 complex, thereby driving NF-kappa-B activation and pro-inflammatory cytokine secretion via MyD88-, TIRAP- and TRAF6-dependent pathways. Related references: PMID:20133493, PMID:22265692, PMID:23264655, PMID:8612135
  • Additionally, CD14 acts as a coreceptor for TLR2:TLR6 and TLR2:TLR1 heterodimers, recognizing diacylated and triacylated lipopeptides, respectively, and triggering lipid-raft-dependent signaling. Related references: PMID:16880211
  • CD14 also binds electronegative LDL (LDL(-)) and mediates the cytokine release induced by LDL(-), implicating it in atherosclerosis-related inflammation. Related references: PMID:23880187
  • At the protein level, CD14 is strongly detected on the surface of monocytes and weakly on granulocytes; it is also expressed by most tissue macrophages. Related references: PMID:1698311
  • Subcellularly, CD14 is located at the cell membrane, enriched in membrane rafts, and also found in the Golgi apparatus; a secreted form exists as well.
  • The protein is a GPI-anchored glycoprotein with multiple leucine-rich repeats and disulfide bonds, features that are critical for its function.

Experimental Guidance and Technical Tips

  • For flow cytometry staining, titrate the -conjugated antibody using human peripheral blood mononuclear cells (PBMCs) or cynomolgus monkey leukocytes to determine the optimal dilution.
  • Since CD14 is a GPI-anchored membrane protein, use a cell surface staining protocol; avoid permeabilization unless intracellular staining is intended, as this may disrupt raft-localized complexes.
  • Consider co-staining with monocyte/macrophage markers (e.g., CD16, CD163) and viability dyes to accurately gate on monocyte populations.
  • Verify cross-reactivity in cynomolgus samples if advancing to non-human primate studies; the antibody’s reported reactivity with cynomolgus allows translational research.
  • As CD14 can be shed as a soluble form, avoid excessive washing steps that might remove loosely bound antibody; use 0.5% BSA/PBS buffer for staining.
  • For data analysis, use appropriate compensation controls when using , and gate on FSC/SSC to identify monocyte/macrophage populations.
