Biological Background: CD14 Function and Localization
- Monocyte differentiation antigen CD14, also known as My23 antigen or myeloid cell-specific leucine-rich glycoprotein, is a GPI-anchored glycoprotein encoded by the CD14 gene.
- Functions as a coreceptor for bacterial lipopolysaccharide (LPS); in concert with LBP, binds monomeric LPS and delivers it to the LY96/TLR4 complex, triggering innate immune responses. Related references: PMID:1698311, PMID:23264655, PMID:20133493, PMID:22265692
- Acts as a coreceptor for TLR2:TLR6 and TLR2:TLR1 heterodimers in response to diacylated and triacylated lipopeptides, respectively, with signaling from the cell surface and subsequent trafficking to the Golgi via lipid rafts. Related references: PMID:16880211
- Mediates cytokine release induced by electronegative LDL (LDL(-)) through an alternative inflammatory pathway. Related references: PMID:23880187
- Subcellular localization includes the cell membrane, membrane rafts, Golgi apparatus, and a secreted soluble form.
- Highly expressed on the surface of monocytes, macrophages, and weakly on granulocytes; detected on tissue macrophages.
- Key structural features include an N-terminal signal peptide, multiple leucine-rich repeats, a GPI anchor attachment site, and disulfide bonds, essential for proper folding and function.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 20-352 of human CD14, covering the extracellular leucine-rich repeat domain. This design suggests specificity for the mature, surface‑exposed region and should not cross‑react with the signal peptide.
- Since the antibody is directly conjugated to , it is optimized for flow cytometry. Consider titrating the antibody to determine the optimal concentration for your cell type, and include appropriate isotype controls.
- For detection of CD14 on cynomolgus monkey cells, validate reactivity under your specific experimental conditions, as cross‑reactivity may vary with sample preparation.
- Soluble CD14 present in serum or culture supernatant may compete with membrane‑bound CD14; therefore, washing cells thoroughly before staining is recommended.
