Biological Background: CD127/IL-7Rα Function and Localization
- Interleukin-7 receptor subunit alpha (CD127/CDw127) is a type I transmembrane glycoprotein encoded by the IL7R gene.
- It serves as the receptor for interleukin-7 (IL-7), and also binds thymic stromal lymphopoietin (TSLP), playing critical roles in lymphocyte development and homeostasis.
- CD127 belongs to the type I cytokine receptor family and forms heterodimers with the common gamma chain (γc) for IL-7 signaling, and with TSLP receptor for TSLP signaling.
- The protein is primarily localized at the cell membrane, but a secreted isoform exists through alternative splicing.
- Post-translational modifications include N-linked glycosylation, disulfide bond formation, phosphorylation, and ubiquitination.
- Mutations in IL7R are associated with severe combined immunodeficiency (SCID) and other immunological disorders.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 21-236) encompasses the extracellular domain; the resulting antibody is likely to recognize native surface-expressed CD127/IL-7Rα.
- For flow cytometry, consider titrating the antibody on your target cell population to determine the optimal staining concentration.
- Verify cross-reactivity in your specific experimental model; reactivity is confirmed for human and cynomolgus monkey.
- Use appropriate isotype controls and Fc receptor blocking to reduce nonspecific binding.
