Biological Background: β2 Microglobulin Function and Localization

  • Beta-2-microglobulin (B2M) is a 13 kDa non-glycosylated protein that serves as the invariant light chain of MHC class I molecules, essential for antigen presentation to cytotoxic T cells.
  • As a member of the immunoglobulin superfamily, B2M contains an immunoglobulin-like domain and is non-covalently associated with the alpha chain of MHC I on the cell surface.
  • B2M is synthesized as a pre-protein, processed into the mature form upon cleavage of the signal peptide, and is constitutively expressed in various tissues, reflecting its fundamental role in immune surveillance.
  • Subcellularly, B2M is predominantly located on the cell surface as part of the MHC I complex and is also found in secreted form in body fluids.
  • The protein undergoes post-translational modifications including glycation, disulfide bond formation, and pyrrolidone carboxylic acid formation at the N-terminus, which are important for its stability and function.
  • Exogenously applied M. tuberculosis EsxA or EsxA-EsxB binds B2M and impairs its export to the cell surface, potentially compromising class I antigen presentation, which may represent an immune evasion mechanism. Related references: PMID:25356553
  • Mutations in the B2M gene have been linked to amyloidosis and other diseases, highlighting its clinical significance in protein aggregation disorders.

Experimental Guidance and Technical Tips

  • The antibody is generated against a recombinant immunogen spanning amino acids 21-119 of human B2M, encompassing the mature extracellular domain; thus, it likely recognizes the native conformation on live cells, making it suitable for flow cytometry.
  • For flow cytometric analysis of cell surface β2-microglobulin, consider using this conjugate directly diluted in appropriate buffer; a standard staining protocol for human peripheral blood mononuclear cells or lymphocyte cell lines may be used.
  • Validate the optimal antibody concentration by titration in the relevant experimental system, and include appropriate isotype controls to ensure specificity.
  • As B2M is a soluble factor, supernatants or plasma may be analyzed using bead-based flow assays; however, the performance of this antibody in such assays has not been specifically tested.
