Biological Background: ULBP2 Function and Localization
- ULBP2 (UL16-binding protein 2) is also known as ALCAN-alpha, NKG2D ligand 2 (NKG2DL2), and retinoic acid early transcript 1H.
- It functions as a ligand for the KLRK1/NKG2D receptor, an activating receptor on natural killer (NK) cells, mediating NK cell cytotoxicity.
- ULBP2 is a member of the RAET1 family of NKG2D ligands, which play critical roles in immune surveillance against infected and transformed cells.
- Subcellularly, ULBP2 localizes to the cell membrane, endoplasmic reticulum, and can be secreted.
- It is attached to the membrane via a GPI-anchor and contains disulfide bonds and glycosylation sites.
- Expression is largely restricted to fetal tissues and various cancer cell lines, with minimal expression in normal adult tissues, highlighting its potential as a tumor-associated antigen.
- ULBP2 is involved in host-virus interactions, as certain viruses have evolved mechanisms to downregulate NKG2D ligands to evade immune detection.
Experimental Guidance and Technical Tips
- For flow cytometry, titrate the antibody to determine the optimal concentration for your specific cell type and experimental conditions.
- Include appropriate isotype controls and viability dyes to ensure accurate gating and detection of ULBP2 surface expression.
- Validate antigen expression in relevant cancer cell lines or activated immune cells, as ULBP2 is not typically expressed in normal resting tissues.
- The immunogen corresponds to the extracellular domain (aa 26-217), so the antibody is expected to recognize native ULBP2 on live, unfixed cells.
