- ULBP2 (UL16-binding protein 2), also known as ALCAN-alpha, N2DL2, NKG2DL2, or RAET1H, is a GPI-anchored membrane protein and a member of the RAET1 family of NKG2D ligands.
- It serves as a ligand for the KLRK1/NKG2D receptor, an activating receptor expressed on natural killer (NK) cells and some T cell subsets, triggering cytotoxic immune responses.
- Upon engagement with NKG2D, ULBP2 mediates NK cell-mediated cytotoxicity against target cells, playing a critical role in tumor immunosurveillance.
- ULBP2 is localized on the cell membrane via a GPI anchor, but a secreted form also exists; it is additionally detected in the endoplasmic reticulum.
- Its expression is highly tissue-restricted: it is found in various cancer cell lines and fetal tissues, but is absent in normal adult tissues, highlighting its potential as a tumor-associated antigen.
