Biological Background: TREM1 Function and Localization

  • TREM1 (Triggering receptor expressed on myeloid cells 1), also designated CD354 or TREM-1, is a cell surface receptor of the immunoglobulin superfamily that amplifies inflammatory responses upon ligand binding by forming a complex with TYROBP/DAP12.   • Upon activation, TREM1 triggers a SYK-mediated tyrosine phosphorylation cascade, leading to downstream activation of BTK, MAPK1, MAPK3, and phospholipase C-gamma, which drives pro-inflammatory cytokine and chemokine release. Related references: PMID:14656437, PMID:21659545
  • TREM1 amplifies inflammatory signals initiated by Toll-like receptor (TLR) and NOD-like receptor engagement, playing a key role in septic shock and atherosclerosis pathophysiology. Related references: PMID:11323674, PMID:21393102
