Biological Background: TNFRSF10D/TRAILR4 Function and Localization
- TNFRSF10D, also known as decoy receptor 2 (DcR2), TRAIL-R4, or CD264, is a member of the tumor necrosis factor receptor superfamily.
- It functions as a receptor for the cytotoxic ligand TRAIL but contains a truncated death domain, rendering it incapable of inducing apoptosis; instead, it protects cells against TRAIL-mediated apoptosis. Related references: PMID:9430226 PMID:9537512
- Reports are contradictory regarding its ability to induce the NF-kappa-B pathway; some studies suggest it can, while others indicate it cannot. Related references: PMID:9382840 PMID:9430226
- The protein is widely expressed, with highest levels in fetal kidney, lung, and liver, and in adult testis and liver; it is also found in peripheral blood leukocytes and various other tissues.
- Subcellularly, it is a membrane protein, predicted to have a signal peptide, extracellular domain, transmembrane helix, and a truncated intracellular death domain.
- Post-translational modifications include glycosylation and disulfide bond formation, as annotated in UniProt.
- It belongs to the TRAIL receptor family, which includes pro-apoptotic receptors (TRAIL-R1, TRAIL-R2) and other decoy receptors (TRAIL-R3, osteoprotegerin).
Experimental Guidance and Technical Tips
- This antibody is validated for flow cytometry; for optimal performance, titrate the antibody on your target cell type to determine the best working concentration.
- The immunogen corresponds to the extracellular domain of TRAILR4 (aa 56-211), ensuring recognition of the native surface receptor.
- Consider using appropriate isotype controls and viability dyes in FC experiments to ensure specific staining.
- For detection of TRAILR4 expression, cells may need activation or specific culture conditions to upregulate the receptor, depending on the system.
- Avoid using this antibody for applications beyond FC without independent validation.
