Biological Background: TIM-3/HAVCR2 Function and Localization

  • TIM-3/HAVCR2 (hepatitis A virus cellular receptor 2, also known as CD366, KIM-3, TIMD3) is a cell surface receptor of the immunoglobulin superfamily that modulates innate and adaptive immune responses, with both inhibitory and stimulatory functions depending on cellular context (PubMed:24825777). Related references: PMID:24825777
  • It negatively regulates T-helper type 1 (Th1) lymphocyte-mediated auto- and alloimmune responses and promotes immunological tolerance (PubMed:14556005). In CD8+ T cells, it attenuates TCR-induced signaling by blocking NF-κB and NFAT promoter activities, leading to loss of IL-2 secretion, potentially via recruitment of LCK and PTPRC (PubMed:24337741, PubMed:26492563). Related references: PMID:14556005, PMID:24337741, PMID:26492563
  • TIM-3 acts as a receptor for galectin-9 (LGALS9), and this interaction is proposed to suppress T-cell responses and induce apoptosis of antigen-specific T cells, involving phosphorylation of HAVCR2 and disruption of its interaction with BAG6 (PubMed:16286920, PubMed:24337741). However, the LGALS9 binding function has been challenged (PubMed:23555261). Related references: PMID:16286920, PMID:24337741, PMID:23555261
  • It also functions as a phosphatidylserine (PtSer) receptor, mediating phagocytosis of apoptotic cells by dendritic cells and promoting innate immune responses (By similarity).
  • Subcellularly, TIM-3 localizes to the cell membrane and cell junctions (By similarity).
  • Expression is prominent in Th1 lymphocytes, regulatory T (Treg) cells after TCR stimulation, dendritic cells, natural killer (NK) cells, and epithelial tissues. It is upregulated in chronic viral infections such as hepatitis C and HIV-1 (By similarity).
  • Keywords indicate post-translational modifications: glycoprotein, disulfide bond, palmitoylation (lipoprotein), and phosphorylation (phosphoprotein), which may regulate its function and localization.

Experimental Guidance and Technical Tips

  • The immunogen (aa 22–200) covers the extracellular domain of human TIM-3, making this antibody suitable for detecting surface-expressed receptor in flow cytometry.
  • As a directly conjugated antibody (), it streamlines the staining protocol and minimizes background; ensure proper compensation controls in multicolor panels.
  • Validate the antibody in relevant biological samples such as Th1 cells, Tregs, NK cells, or dendritic cells, where TIM-3 expression is well-documented.
  • Consider pairing with viability dyes or other lineage markers to accurately gate TIM-3+ populations, especially in apoptotic cell phagocytosis assays.
