Biological Background: TIM-3/HAVCR2 Function and Localization

  • TIM-3 (HAVCR2) is a T-cell immunoglobulin and mucin domain-containing protein 3, also known as CD366, KIM-3, or TIMD-3. It is a cell surface receptor primarily associated with modulating innate and adaptive immune responses. Related references: PMID:11823861
  • It generally exerts inhibitory functions, suppressing Th1-mediated auto- and alloimmune responses and promoting immunological tolerance.   • TIM-3 is expressed on various immune cells, including Th1 lymphocytes, Treg cells, dendritic cells, and natural killer cells. Experimental Guidance and Technical Tips

  • This antibody is generated against the extracellular domain (aa 22–200) of human TIM-3, providing specificity for the native conformation on live cells. Consider using it for surface staining in flow cytometry without permeabilization.
  • For flow cytometry, titrate the antibody to determine the optimal staining concentration for your specific cell type and experimental conditions. Include appropriate isotype controls and Fc receptor blocking steps.
  • Although the antibody is validated for flow cytometry, cross-reactivity with other species has not been tested; its use is restricted to human samples. Validate specificity using TIM-3 overexpressing cells or knockdown/knockout models.
