Biological Background: Tie-2/CD202b (Angiopoietin-1 receptor) Function and Localization

  • Tie-2/CD202b (Angiopoietin-1 receptor) is a tyrosine-protein kinase receptor encoded by the TEK gene, also known as endothelial tyrosine kinase, Tunica interna endothelial cell kinase, and p140 TEK.
  • It serves as a cell-surface receptor for angiopoietins (ANGPT1, ANGPT2, and ANGPT4), playing a critical role in regulating angiogenesis, endothelial cell survival, proliferation, migration, adhesion, and vascular quiescence.
  • The receptor is essential for normal vascular development and heart formation during embryogenesis, as well as for postnatal hematopoiesis.
  • Tie-2 modulates both angiogenesis activation and inhibition depending on the cellular context: in quiescent vessels, it promotes vascular stability via ANGPT1 oligomers recruiting TEK to cell-cell contacts and activating PI3K/AKT signaling; in migrating endothelial cells, it stimulates sprouting angiogenesis through focal adhesion complex formation and MAPK/ERK pathway activation.
  • Subcellularly, Tie-2 is primarily located at the cell membrane, but also found at cell junctions, focal adhesions, within the cytoplasm, and as a secreted soluble ectodomain generated by proteolytic processing.
  • Predominantly expressed in endothelial cells and their progenitors, with high levels detected in placenta and lung and lower levels in brain, kidney, and umbilical vein endothelial cells.
  • Key structural features include immunoglobulin-like domains, EGF-like domains, a transmembrane segment, and a cytoplasmic tyrosine kinase domain; the receptor is subject to autophosphorylation and ubiquitination.

Experimental Guidance and Technical Tips

  • Consider titrating the antibody on human endothelial cells (e.g., HUVECs or EA.hy926) to determine the optimal concentration for flow cytometry.
  • Validate antibody performance in the relevant sample system, ensuring that the cells express Tie-2/CD202b.
  • For multicolor flow panels, include proper compensation controls using cells or beads stained with each fluorophore individually.
  • Note that soluble Tie-2 extracellular domain may be present in serum or plasma and could potentially interfere with staining; appropriate blocking steps or validation in the intended sample matrix is advised.
