- Tie-2/CD202b (Angiopoietin-1 receptor) is a tyrosine-protein kinase receptor encoded by the TEK gene, also known as endothelial tyrosine kinase, Tunica interna endothelial cell kinase, and p140 TEK.
- It serves as a cell-surface receptor for angiopoietins (ANGPT1, ANGPT2, and ANGPT4), playing a critical role in regulating angiogenesis, endothelial cell survival, proliferation, migration, adhesion, and vascular quiescence.
- The receptor is essential for normal vascular development and heart formation during embryogenesis, as well as for postnatal hematopoiesis.
- Tie-2 modulates both angiogenesis activation and inhibition depending on the cellular context: in quiescent vessels, it promotes vascular stability via ANGPT1 oligomers recruiting TEK to cell-cell contacts and activating PI3K/AKT signaling; in migrating endothelial cells, it stimulates sprouting angiogenesis through focal adhesion complex formation and MAPK/ERK pathway activation.
- Subcellularly, Tie-2 is primarily located at the cell membrane, but also found at cell junctions, focal adhesions, within the cytoplasm, and as a secreted soluble ectodomain generated by proteolytic processing.
- Predominantly expressed in endothelial cells and their progenitors, with high levels detected in placenta and lung and lower levels in brain, kidney, and umbilical vein endothelial cells.
- Key structural features include immunoglobulin-like domains, EGF-like domains, a transmembrane segment, and a cytoplasmic tyrosine kinase domain; the receptor is subject to autophosphorylation and ubiquitination.
