Biological Background: T-bet/Tbx21 Function and Localization
- T-bet (TBX21) is a T-box transcription factor, also known as T-cell-specific T-box transcription factor T-bet, T-PET, TBET, or TBLYM, and is the master regulator of Th1 lineage commitment.
- Drives Th1 differentiation by activating Th1 genetic programs (e.g., IFNG, CXCR3) and repressing Th2/Th17 programs; suppresses Th2 cytokines (IL-4, IL-5, IL-13) via repression of GATA3 and NFATC2, and inhibits Th17 by blocking RUNX1-mediated RORC transactivation. Related references: PMID:10761931
- Epigenetically remodels the IFNG locus by inducing permissive chromatin accessibility and CpG methylation, recruiting chromatin remodeling complexes including KDM6B, SWI/SNF, and an H3K4me2-methyltransferase complex. Related references: PMID:33296702

- This antibody is validated for intracellular flow cytometry (FC intra). For optimal detection of nuclear T-bet, use fixation and permeabilization buffers optimized for nuclear antigens, and titrate the antibody to minimize background.
- Include appropriate isotype controls and compare resting vs. activated T-cell populations to confirm specificity, as T-bet expression is activation-dependent.
